Biological Background: SDHA Function and Localization
- SDHA encodes the flavoprotein subunit of succinate dehydrogenase (complex II), which oxidizes succinate to fumarate in the tricarboxylic acid cycle and transfers electrons to ubiquinone in the mitochondrial electron transport chain. Related references: PMID:10746566 PMID:24781757
- Additionally, SDHA can oxidize malate to enol-oxaloacetate, which is a potent inhibitor of succinate dehydrogenase; the enol form is then isomerized to keto-oxaloacetate.
- SDHA acts as a tumor suppressor, with loss-of-function mutations linked to paraganglioma and pheochromocytoma. Related references: PMID:20484225
- The protein localizes to the mitochondrial inner membrane, contains a FAD cofactor, and undergoes post-translational modifications including acetylation and phosphorylation.
- Alternative splicing generates multiple isoforms, and mutations in SDHA are associated with Leigh syndrome, cardiomyopathy (CMD1GG), and neurodegeneration.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 385-664 (C-terminal region); the antibody may recognize full-length SDHA at the expected molecular weight of ~73 kDa in WB using mitochondrial lysates or whole-cell extracts.
- For IHC-P and IF/ICC, anticipate mitochondrial staining; validate in tissue/cell types known to express SDHA, such as skeletal muscle, heart, or kidney.
- Since cross-reactivity is reported for human, mouse, and rat, confirm reactivity in the relevant model system and optimize detection conditions.
- In ELISA, ensure sufficient target protein availability; recombinant SDHA protein or lysates from expressing cells can serve as positive controls.
