Biological Background: SCYL2 Function and Localization
- SCYL2 (SCY1-like protein 2), also known as coated vesicle-associated kinase of 104 kDa (CVAK104), is a member of the SCY1-like family of protein kinases.
- It is a component of the AP2-containing clathrin coat and is involved in regulating clathrin-dependent trafficking at the plasma membrane, trans-Golgi network (TGN), and endosomal system (Probable).
- SCYL2 has been proposed to act as a serine/threonine-protein kinase targeting the beta2-subunit of the AP2 adapter complex and other proteins in the presence of poly-L-lysine, though this activity remains unconfirmed. Related references: PMID:15809293 PMID:16914521
- By modulating the expression of excitatory receptors at synapses, SCYL2 plays an essential role in neuronal function, signaling, and brain development (By similarity).
- The protein localizes to cytoplasmic vesicles, specifically clathrin-coated vesicles, as well as to the trans-Golgi network membrane and endosome membrane.
- Post-translational modifications include phosphorylation; SCYL2 is classified as a phosphoprotein.
- Keywords associated with SCYL2 include Coiled coil, Cytoplasmic vesicle, Disease variant, Endosome, Golgi apparatus, Membrane, Phosphoprotein, Proteomics identification, and Reference proteome.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 1-300) of human SCYL2, allowing detection of endogenous protein across human, mouse, and rat samples.
- For Western blot (WB), consider using brain tissue or neuronal cell lines as positive controls, given SCYL2's role in neuronal function.
- In immunofluorescence/immunocytochemistry (IF/ICC), anticipate staining patterns consistent with clathrin-coated vesicles, TGN, and endosomal compartments.
- For ELISA, the recombinant immunogen can serve as a positive control; validate assay conditions in the relevant sample matrix.
