Biological Background: SCYL2 Function and Localization
- SCYL2 (SCY1-like protein 2, also known as coated vesicle-associated kinase of 104 kDa/CVAK104) is a component of the AP2-containing clathrin coat, likely regulating clathrin-dependent trafficking at the plasma membrane, trans-Golgi network (TGN), and endosomal system.
- A possible serine/threonine-protein kinase activity toward the beta2-subunit of the AP2 adaptor complex and other substrates in the presence of poly-L-lysine has been reported but remains unconfirmed. Related references: PMID:15809293 PMID:16914521
- By regulating synaptic excitatory receptor expression, SCYL2 plays an essential role in neuronal function, signaling, and brain development (by similarity).
- Localizes to clathrin-coated vesicles, trans-Golgi network membrane, and endosome membranes.
- Belongs to the SCY1-like family, contains coiled-coil domains, and is a phosphoprotein identified in proteomics studies; mutations are linked to disease variants.
Experimental Guidance and Technical Tips
- This antibody was generated against an N-terminal fragment (aa 1–300) of human SCYL2 and is suitable for western blot (WB) and ELISA applications.
- For WB, consider using reducing conditions and validate the ~104 kDa band in the appropriate lysate; optimization of primary antibody dilution may be necessary.
- With reported cross-reactivity to rat, the antibody may be useful for neuronal tissue studies in this species, but species-specific reactivity should be verified in your assay system.
- Given SCYL2’s roles in vesicle trafficking and neuronal signaling, testing in brain or neuronal cell line lysates is recommended for functional studies.
