Biological Background: SCP2 Function and Localization
- SCP2 encodes isoforms SCP2 (sterol carrier protein 2) and SCPx (sterol carrier protein X/SCP-X), also known as non-specific lipid-transfer protein (NSL-TP), acetyl-CoA C-myristoyltransferase, and propanoyl-CoA C-acyltransferase.
- It catalyzes the last step of peroxisomal beta-oxidation of branched-chain fatty acids and bile acid intermediates, and is active with medium and long straight chain 3-oxoacyl-CoAs. Related references: PMID:10706581
- SCP2 mediates the transfer of phospholipids, cholesterol, and gangliosides between membranes, and stimulates the conversion of 7-dehydrocholesterol to cholesterol, thereby playing a role in regulating steroidogenesis. Related references: PMID:17157249 PMID:7642518 PMID:8300590
- The protein localizes to peroxisomes, cytoplasm, mitochondria, and endoplasmic reticulum, consistent with its roles in lipid metabolism and transport.
- SCP2 is expressed predominantly in liver, fibroblasts, and placenta.
- Post-translational modifications include acetylation and phosphorylation, and the protein is identified in direct protein sequencing and proteomics identification studies.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the full-length SCP2 isoform (1-322), making this antibody suitable for detecting both SCP2 and potentially SCPx if the epitope is conserved; however, isoform-specific validation is recommended.
- For Western blot (WB), the predicted molecular weight is 59 kDa (full-length, possibly SCPx), but SCP2 isoform may appear smaller; validate band sizes in the relevant sample system.
- For immunohistochemistry (IHC-P), consider antigen retrieval methods and include peroxisome-rich tissues (e.g., liver) as positive controls.
- For ELISA, ensure the coating antigen matches the immunogen region and optimize the antibody dilution.
- Since the antibody cross-reacts with mouse, it can be used for comparative studies in murine models; however, confirm specificity in genetically modified controls if possible.
