Biological Background: SCO1 Function and Localization
- SCO1 is a copper metallochaperone essential for the maturation of cytochrome c oxidase subunit II (MT-CO2/COX2) by delivering copper to the Cu(A) site, in cooperation with SCO2. Related references: PMID:15229189 PMID:15659396 PMID:16735468
- SCO1 also plays a role in regulating copper homeostasis by controlling the abundance and cell membrane localization of copper transporter CTR1 (By similarity).
- The protein is localized to the mitochondrial inner membrane.
- It is predominantly expressed in tissues with high oxidative phosphorylation, such as muscle, heart, and brain.
- Mutations in SCO1 are associated with primary mitochondrial disease (MC4DN4); the protein contains a disulfide bond, a transmembrane helix, and a copper-binding domain.
