Biological Background: SCN5A Function and Localization

  • SCN5A encodes the pore-forming alpha subunit of the voltage-gated sodium channel Nav1.5, which mediates the fast depolarizing Na+ current essential for action potential generation in excitable cells. Related references: PMID:33712541, PMID:40768348

  • Nav1.5 is the predominant sodium channel in cardiomyocytes, responsible for the rapid upstroke of the cardiac action potential and thus initiation of the heartbeat. Related references: PMID:11234013, PMID:11804990, PMID:12569159

  • Voltage-dependent gating allows selective Na+ influx upon membrane depolarization, propagating electrical signals throughout tissues. Related references: PMID:1309946, PMID:21447824, PMID:23085483

  • The channel localizes primarily to the sarcolemma and T-tubules of cardiac myocytes, with additional perinuclear and cell junction distribution.

  • SCN5A is predominantly expressed in cardiac muscle; it is also detected at lower levels in jejunal smooth muscle and, as an alternatively spliced isoform, in brain.

  • The Nav1.5 protein undergoes extensive post-translational modifications including phosphorylation, ubiquitination, and methylation, and mutations are linked to cardiac arrhythmias such as Brugada syndrome, long QT syndrome type 3, and atrial fibrillation.

Experimental Guidance and Technical Tips

  • This polyclonal antibody was raised against a C‑terminal peptide (aa 1950‑2016) and is suitable for Western blot detection of full‑length Nav1.5 (~227 kDa).
  • For ELISA applications, consider using the immunizing peptide as a positive control.
  • Cross‑reactivity with mouse has been reported; validate reactivity in the relevant mouse tissue given potential sequence differences in the immunogen region.
