Biological Background: Scinderin (SCIN) Function and Localization
- Scinderin (also known as Adseverin) is a Ca²⁺-dependent actin filament-severing protein that regulates exocytosis by modulating the cortical actin network beneath the plasma membrane. Related references: PMID:26365202 PMID:8547642
- It also exhibits barbed end capping and nucleating activities in vitro, and its severing activity is inhibited by phosphatidylinositol 4,5-bisphosphate (PIP2).
- SCIN is required for megakaryocyte differentiation, maturation, polyploidization, and the release of platelet-like particles, and it inhibits cell proliferation and tumorigenesis through MAPK, p38, and JNK signaling pathways. Related references: PMID:11568009
- Subcellularly, SCIN localizes to the cytoplasm, cytoskeleton, and podosomes (cell projections).
- It is predominantly expressed in megakaryocytes.
