- Secretogranin-2 (SCG2), also designated Chromogranin-C or Secretogranin II (SgII), is a member of the granin family of neuroendocrine secretory proteins.
- It functions in the regulation of secretory granule biogenesis, including the sorting of cargo and formation of dense-core vesicles.
- SCG2 is a secreted protein localized to secretory granules and is released upon stimulation.
- The protein undergoes post-translational modifications such as glycosylation, phosphorylation, sulfation, and proteolytic cleavage at paired basic residues.
- SCG2 contains calcium-binding domains, which contribute to its role in granule stability and aggregation.
Experimental Guidance and Technical Tips
- This polyclonal antibody, generated against amino acids 30-320, may recognize both full-length and processed forms of SCG2; include appropriate controls in your assays.
- For Western blotting, the predicted molecular weight of full-length SCG2 is approximately 71 kDa; additional bands from cleavage products (e.g., EM66) may be observed depending on the sample.
- In immunofluorescence (IF-P), intracellular detection of SCG2 in secretory granules requires cell permeabilization.
- The antibody has been tested in ELISA and can be used for quantitative detection of SCG2 in cell supernatants or lysates.
- Given cross-reactivity with rat, validate the antibody in rat-derived samples using positive control tissues such as pituitary or neuroendocrine cells.
