Biological Background: SCD5 Function and Localization
- SCD5 encodes stearoyl-CoA desaturase 5, an enzyme that introduces the first double bond into saturated fatty acyl-CoA substrates, catalyzing the insertion of a cis double bond at the delta-9 position. Related references: PMID:15610069, PMID:15907797, PMID:22745828
- It generates a mixture of 16:1 and 18:1 unsaturated fatty acids. Related references: PMID:15610069, PMID:15907797
- SCD5 is involved in neuronal cell proliferation and differentiation through down-regulation of EGFR/AKT/MAPK and Wnt signaling pathways. Related references: PMID:22745828
- The protein localizes to the endoplasmic reticulum membrane.
- It is expressed in fetal brain, adult brain, pancreas, and at lower levels in kidney and lung; also detected in spiral ganglion cells and organ of Corti of fetal cochlea. Related references: PMID:31972369
- SCD5 belongs to the fatty acid desaturase family and is also known as SCD4, ACOD4, FADS4, HSCD5, DFNA79.
- Mutations in SCD5 are associated with non-syndromic deafness (DFNA79), and the protein may play a role in hearing.
- It requires iron as a cofactor and contains transmembrane helices.
Experimental Guidance and Technical Tips
- The immunogen was designed from a synthetic peptide spanning amino acids 231-330 of human SCD5, which may provide target specificity; consider validating in the relevant sample system.
- For Western blot, the predicted molecular weight is 38 kDa; include appropriate positive controls (e.g., human, mouse, or rat tissue lysates) and optimize loading amounts to account for species-specific expression levels.
- In ELISA assays, titrate the antibody and use suitable coating buffers; cross-reactivity with mouse and rat SCD5 may be exploited for multi-species studies.
- Due to alternative splicing and potential isoforms, confirm band patterns with reference samples and consider additional validation methods if needed.
