Biological Background: SCD1 Function and Localization
- SCD1, also known as acyl-CoA desaturase or delta-9 desaturase, is a key enzyme in fatty acid metabolism encoded by the SCD gene.
- Catalyzes the insertion of a cis double bond at the delta-9 position of palmitoyl-CoA and stearoyl-CoA, utilizing O2 and cytochrome b5 electrons, to produce 16:1 and 18:1 monounsaturated fatty acids. Related references: PMID:15907797 PMID:18765284 PMID:15610069
- Plays a critical role in lipid biosynthesis, contributing to membrane phospholipids, cholesterol esters, and triglycerides, and regulating lipogenic gene expression and mitochondrial fatty acid oxidation. Important for body energy homeostasis.

- Highly expressed in adult adipose tissue; detected in fetal liver, lung, and brain, and at lower levels in adult brain and lung.

- In immunohistochemistry (paraffin), optimize antigen retrieval; positive staining is anticipated in adipose and hepatic tissues.
- For immunofluorescence/ICC, permeabilization is necessary to detect the ER-membrane localized SCD protein; include appropriate controls.
- In ELISA applications, consider using recombinant SCD or the immunizing peptide as a positive control to confirm binding.
- Cross-reactivity with mouse and rat suggests the epitope is conserved; this may facilitate translational studies in rodent models.
