Biological Background: Stearoyl-CoA desaturase (SCD) Function and Localization
- Stearoyl-CoA desaturase (SCD), also known as Delta-9 desaturase, catalyzes the insertion of a cis double bond at the delta-9 position of fatty acyl-CoA substrates, requiring molecular oxygen and electrons from reduced cytochrome b5. Related references: PMID:15907797, PMID:18765284
- The enzyme converts palmitoyl-CoA (16:0) and stearoyl-CoA (18:0) into palmitoleate (16:1) and oleate (18:1), respectively, generating a mixture of monounsaturated fatty acids. Related references: PMID:15610069
- SCD plays a central role in lipid biosynthesis, regulating lipogenic gene expression and mitochondrial fatty acid oxidation, thereby contributing to whole-body energy homeostasis.
- It is essential for the biosynthesis of membrane phospholipids, cholesterol esters, and triglycerides.
- Subcellularly, SCD localizes to the endoplasmic reticulum membrane as a multi-pass transmembrane protein.
- Tissue expression is highest in adult adipose tissue; moderate levels are found in fetal liver, lung, and brain, with lower expression in adult brain and lung.
- The protein contains iron-binding sites critical for its desaturase activity and is identified as a phosphoprotein in proteomics studies.

- The immunogen is a synthetic peptide derived from amino acids 260–359 of human SCD. For Western blot, a band at approximately 42 kDa is expected; consider using appropriate molecular weight markers.
- In immunofluorescence/immunocytochemistry (IF/ICC), optimize fixation and permeabilization conditions to ensure detection of the ER-membrane-localized target.
