Biological Background: SCAF8 Function and Localization
- SCAF8 (SR-related and CTD-associated factor 8), also known as RBM16 or CCAP7, is a 141 kDa nuclear protein that functions as an anti-terminator during transcription.
- Together with SCAF4, it prevents early mRNA termination by suppressing the use of alternative polyadenylation sites, thereby ensuring full-length transcript synthesis. Related references: PMID:31104839
- SCAF8 associates with the phosphorylated C-terminal domain (CTD) of RNA polymerase II and binds nascent RNA upstream of early poly(A) sites to inhibit premature cleavage and polyadenylation. Related references: PMID:31104839

- Subcellularly, SCAF8 is localized to the nucleus and nuclear matrix, consistent with its role in co-transcriptional RNA processing.

- The immunogen is a recombinant fusion protein spanning amino acids 572–634 of human SCAF8. This region is expected to be immunogenic; however, users should validate antibody specificity in their target species and sample type.
- For Western blotting, SCAF8 has a predicted molecular weight of 141 kDa. We recommend using a low-percentage polyacrylamide gel (e.g., 6–8%) and extended transfer times to ensure efficient transfer of this high-molecular-weight protein.
- In ELISA applications, optimization of coating antigen and antibody dilution is advised. Consider using recombinant SCAF8 or cell lysates as positive controls.
- This polyclonal antibody is reported to cross-react with mouse and rat SCAF8. Users should verify reactivity if using samples from other species.
