- Protein salvador homolog 1 (SAV1), also known as 45 kDa WW domain protein (hWW45), is a key scaffolding component of the Hippo signaling pathway.
- SAV1 functions as a regulator of STK3/MST2 and STK4/MST1 kinases, forming a complex that phosphorylates and activates LATS1/2, leading to YAP1 inactivation. This cascade restricts proliferation and promotes apoptosis, playing a pivotal role in organ size control and tumor suppression. Related references: PMID:29063833
- It promotes cell-cycle exit and terminal differentiation in developing epithelial tissues.
- SAV1 regulates centrosome disjunction by modulating NEK2 localization and its phosphorylation of CROCC and CEP250.
- In conjunction with STK3/MST2, SAV1 can activate transcriptional activity of estrogen receptor alpha (ESR1) through phosphorylation modulation.
