Biological Background: SAV1 Function and Localization

  • SAV1 (Protein salvador homolog 1, also known as hWW45) is a 45 kDa WW domain-containing scaffold protein and a core component of the Hippo signaling pathway, which controls organ size and suppresses tumorigenesis by coupling cell proliferation to apoptosis (PubMed:29063833). Related references: PMID:29063833
  • SAV1 forms a complex with the kinases STK3/MST2 and STK4/MST1, facilitating their activation and subsequent phosphorylation of the LATS1/2-MOB1 complex, which in turn phosphorylates and retains the oncogenic transcriptional co-activators YAP1 and WWTR1/TAZ in the cytoplasm, preventing their nuclear entry and pro-growth gene expression.
  • Beyond canonical Hippo signaling, SAV1 regulates centrosome disjunction by controlling the localization of NEK2 to centrosomes, influencing the phosphorylation of CROCC and CEP250, and also modulates estrogen receptor alpha (ESR1) transcriptional activity through STK3/MST2-dependent phosphorylation.
  • SAV1 localizes to both the nucleus and cytoplasm, consistent with its dual roles in transcriptional regulation and cytoplasmic kinase scaffold functions.
  • The protein is ubiquitously expressed in adult human tissues, with highest levels in the pancreas, aorta, and interventricular septum, and lowest in skeletal muscle; expression is enhanced in fetal heart compared to adult heart, suggesting developmental roles.
  • Post-translational modifications include phosphorylation, and the protein features a coiled-coil domain and WW domain repeats (keywords: Phosphoprotein, Coiled coil, Repeat, 3D-structure).

Experimental Guidance and Technical Tips

  • This rabbit polyclonal antibody was raised against a recombinant fusion protein encompassing amino acids 199-383 of human SAV1 (NP_068590.1), which covers the C-terminal region containing the coiled-coil domain and WW domain repeats important for protein-protein interactions.
  • For Western blotting, SAV1 has a predicted molecular weight of ~45 kDa; we recommend using appropriate loading controls and confirming the band size in relevant cell lysates (e.g., HeLa, HEK293, or mouse tissue samples).
  • For ELISA, consider optimizing coating antigen concentration and antibody dilution using standard indirect ELISA protocols.
  • Since the antibody cross-reacts with both human and mouse, it is suitable for comparative studies in these species; however, validation in the specific experimental system is advised.
  • Note that SAV1 is expressed in many tissues, so consider using cell lines with known expression levels as positive controls.
