Biological Background: SATB2 Function and Localization
- DNA-binding protein SATB2 (Special AT-rich sequence-binding protein 2) binds to matrix attachment regions (MARs) of DNA, recognizing the sugar-phosphate backbone of double-stranded DNA.
- Acts as a transcription factor that recruits chromatin remodeling enzymes and coregulators (HDACs or HATs) to promoters and enhancers, thereby modulating local chromatin structure and gene expression.
- Plays a critical role in cortical development by initiating upper-layer neuron specification and repressing deep-layer neuron genes such as Ctip2 (BCL11B).
- Involved in palate formation and skeletal development, serving as a molecular node in transcriptional networks that regulate osteoblast differentiation.
- Localizes to the nucleus matrix, consistent with its function in chromatin organization and nuclear architecture.
- Exhibits tissue-specific expression, with high levels in adult brain regions (amygdala, corpus callosum, caudate nucleus, hippocampus), moderate in fetal brain, and low in liver, kidney, and spinal cord.
- Subject to alternative splicing, phosphorylation (phosphoprotein), and ubiquitination (Ubl conjugation), which may modulate its activity and stability (isopeptide bond).
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 200–300 of human SATB2; verify sequence alignment with mouse and rat orthologs to confirm expected cross-reactivity.
- For western blot (WB), a specific band at approximately 83 kDa is anticipated; use reducing sample buffer and include a prestained protein ladder to monitor molecular weight.
- In immunohistochemistry on paraffin-embedded sections (IHC-P), antigen retrieval (e.g., heat-induced with citrate buffer, pH 6.0) may be required; validate staining pattern with appropriate positive and negative control tissues.
- When performing immunoprecipitation (IP), titrate the antibody concentration and include an isotype-matched control IgG to assess specificity.
- For ELISA, pair this antibody with a suitable detection antibody; optimization of coating antigen and antibody concentrations is recommended.
