- SATB2 (Special AT-rich sequence-binding protein 2) is a DNA-binding protein that recognizes the sugar-phosphate backbone of double-stranded DNA and binds to nuclear matrix or scaffold attachment regions (MARs).
- It acts as a transcription factor and chromatin regulator, recruiting both corepressors (HDACs) and coactivators (HATs) to modulate local chromatin-loop remodeling at promoters and enhancers.
- Plays a critical role in cortical development by initiating upper-layer neuron (UL1) genetic programs and repressing deep-layer neuron genes, partly through regulation of BCL11B (Ctip2), thereby influencing corticocortical connectivity.
- Involved in skeletal development and osteoblast differentiation, serving as a molecular node in transcriptional networks governing bone formation.
- May also contribute to palate formation during embryogenesis.
- Localizes to the nuclear matrix, consistent with its role in chromatin organization and gene regulation.
- High expression in adult brain (especially amygdala, corpus callosum, caudate nucleus, hippocampus), moderate in fetal brain, and weak in adult liver, kidney, and spinal cord.
- Post-translationally modified by phosphorylation and ubiquitination; subject to alternative splicing and linked to chromosomal rearrangements implicated in intellectual disability and other diseases.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a region within amino acids 634-733 (NP_001165980.1); consider verifying the sequence conservation in your species of interest when assessing cross-reactivity.
- For Western blotting (WB), a band at approximately 83 kDa is expected; optimize antibody dilution using a positive control such as human or mouse brain lysate.
- For immunofluorescence (IF) and immunohistochemistry (IHC), nuclear localization should be observed; include appropriate nuclear counterstaining and validate in relevant tissue sections, especially brain or bone.
- For immunoprecipitation (IP), consider using a cross-linking method if preserving protein-DNA interactions is desired, given SATB2’s chromatin-binding function.
