- SATB1 (Special AT-rich sequence-binding protein 1) is a DNA-binding protein that specifically recognizes the sugar-phosphate backbone of double-stranded DNA at special AT-rich sequences, predominantly at nuclear matrix/scaffold attachment regions (MARs).
- Acts as a transcriptional repressor or activator by inducing local chromatin-loop remodeling and serving as a docking platform for chromatin-modifying enzymes such as histone acetyltransferases (HATs) and deacetylases (HDACs), thereby regulating gene expression.
- Plays a crucial role in T-cell development by modulating genes essential for the maturation of CD8 single-positive thymocytes.
- Mediates X chromosome inactivation during embryogenesis by acting as a silencing factor downstream of Xist RNA.
- Regulates globin gene switching during erythroid differentiation, controlling fetal and adult hemoglobin expression.
- Facilitates HIV-1 integration by creating a favored chromatin context at Alu-like motifs, and HIV-1 Tat protein can override SATB1-mediated repression of interleukin genes.
- Reprogrammes chromatin organization and transcription in breast tumors, promoting metastasis by upregulating metastasis-associated genes and downregulating tumor suppressors.
