Biological Background: SART3 Function and Localization
- SART3 (Spliceosome associated factor 3), also known as SART-3, Tip110, or p110 nuclear RNA-binding protein, is a 110 kDa U6 snRNP-binding protein crucial for spliceosome recycling.
- Acts as a recycling factor for U4/U6 snRNPs, promoting the reassembly of U4 and U6 snRNPs after spliceosome disassembly, a key step in pre-mRNA splicing. Related references: PMID:12032085
- Also binds U6atac snRNPs and functions in the recycling of U4atac/U6atac snRNPs, facilitating U12-type spliceosome assembly for splicing non-canonical introns. Related references: PMID:14749385

Experimental Guidance and Technical Tips
- The immunogen is a recombinant fragment (aa 664-963), so the antibody may recognize full-length SART3 and any isoforms or degradation products containing this region. Validate specificity in your sample system.
- For Western blot (WB), the predicted molecular weight is ~110 kDa. Note that actual mobility may shift due to post-translational modifications; include appropriate controls.
- In immunohistochemistry (IHC-P), antigen retrieval conditions (e.g., citrate buffer pH 6.0 or Tris-EDTA pH 9.0) should be optimized as nuclear targets can require stronger retrieval.
- For ELISA, use the immunogen fragment as a positive control and determine optimal coating concentration experimentally.
