Biological Background: SARS (Seryl-tRNA Synthetase / SARS1) Function and Localization
- SARS (also designated serine--tRNA ligase, cytoplasmic, Seryl-tRNA synthetase, SerRS) is encoded by the SARS1 gene and belongs to the class II aminoacyl-tRNA synthetase family.
- Catalyzes the ligation of serine to its cognate tRNA(Ser) in a two-step mechanism: serine activation by ATP to form Ser-AMP, followed by transfer to the 3'-end of tRNA(Ser), essential for accurate protein translation. Related references: PMID:22353712 PMID:24095058 PMID:26433229
- Also capable of misacylating tRNA(Sec) with serine to produce L-seryl-tRNA(Sec), the precursor for selenocysteine biosynthesis, thereby contributing to selenoprotein production. Related references: PMID:26433229 PMID:28236339 PMID:34570399
- In the nucleus, directly binds the VEGFA core promoter, blocking MYC-mediated transcriptional activation, and additionally recruits SIRT2 to deacetylate histone H4K16, leading to epigenetic silencing of VEGFA and suppression of angiogenesis. - Localizes to both cytoplasm and nucleus, reflecting its dual roles in translation and transcriptional regulation.

Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal 300 amino acids of human SARS; thus the antibody may detect full-length endogenous protein and potential N-terminal-containing isoforms. Confirm specificity using appropriate controls.
- For Western blotting, expect a band at approximately 59 kDa. Optimize loading and blocking conditions to minimize nonspecific binding typical of polyclonal antibodies.
- In immunohistochemistry (IHC-P), both nuclear and cytoplasmic staining patterns may be observed; validate with known positive/negative tissues such as brain.
- In immunofluorescence/immunocytochemistry (IF/ICC), monitor subcellular localization; consider co-staining with compartment markers.
- For ELISA, titrate antibody and use validated recombinant protein as standard; cross-reactivity with mouse and rat predicted but should be verified experimentally.
