Biological Background: SARS-CoV Spike Function and Localization

  • SARS-CoV Spike glycoprotein (also known as E2 or peplomer protein) is encoded by the S gene and has a molecular weight of approximately 139 kDa. It is a class I viral fusion protein essential for SARS-CoV entry.
  • The protein attaches the virion to host cell receptors, primarily human ACE2 and CLEC4M/DC-SIGNR, initiating infection. Upon receptor binding, the virus is internalized into endosomes.
  • Endosomal acidification triggers cathepsin L (CTSL) proteolysis, which cleaves the Spike protein and unmasks the fusion peptide in the S2 subunit, activating membrane fusion.

  • For Western blot (WB), a band near 139 kDa corresponding to full-length Spike may be observed, and under reducing conditions, S2 fragments may also appear. Consider using appropriate positive controls, such as SARS-CoV-infected cell lysates or recombinant Spike protein.
  • In ELISA applications, the antibody can be used for capture or detection of SARS-CoV Spike protein. Optimize coating and blocking conditions, and validate specificity against other coronaviruses as needed.
  • Since this is a polyclonal antibody, it may exhibit batch-to-batch variability. It is recommended to titrate the antibody for each application and confirm performance in the relevant sample system.
