Biological Background: ORF7a Function and Localization
- ORF7a, also known as Accessory protein 7a, Protein U122, or Protein X4, is a 13744 Da transmembrane protein encoded by SARS-CoV-2.
- It functions as an antagonist of host tetherin (BST2), disrupting its antiviral effect by binding to BST2 and sequestering it to the perinuclear region, thereby preventing BST2 from exerting its antiviral function at the cell membrane. Related references: PMID:33930332
- ORF7a may specifically downregulate the MHC-I allele HLA-A*02:01 (HLA-A2), contributing to viral immune evasion. Related references: PMID:36574644
- Subcellularly, ORF7a localizes to the host cytoplasm, host perinuclear region, and virions, and associates with host endoplasmic reticulum membrane, endoplasmic reticulum-Golgi intermediate compartment membrane, and Golgi apparatus membrane.
- The protein contains a signal peptide, a transmembrane helix, a disulfide bond, and an isopeptide bond; it also undergoes Ubl conjugation.
- Through its interactions, ORF7a inhibits both host innate and adaptive immune responses, including MHC class I molecule presentation and tetherin-mediated restriction.
- ORF7a also modulates host cell cycle by dysregulating the G0/G1 checkpoint.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 16–98 of ORF7a, corresponding to the predicted mature protein after signal peptide cleavage; the antibody is expected to detect the mature form.
- For Western blot, a band at approximately 14 kDa is expected; consider using SARS-CoV-2-infected cell lysates or ORF7a-overexpressing cells as positive controls.
- This antibody is suitable for ELISA; validate performance with recombinant ORF7a protein and relevant biological samples.
- Cross-reactivity with other coronaviruses has not been reported; verify specificity in the experimental system of interest.
