Biological Background: Spike glycoprotein (S) Function and Localization
- Spike glycoprotein (S), also known as E2 or peplomer protein, is encoded by the S gene of SARS-CoV-2 and mediates viral entry into host cells.
- The S protein attaches the virion to the host cell membrane by interacting primarily with the ACE2 receptor (PubMed:32142651, PubMed:32155444, PubMed:33607086). Related references: PMID:32142651 PMID:32155444 PMID:33607086
- Viral entry can also proceed via alternative receptors, including NRP1/NRP2 (PubMed:33082294, PubMed:33082293) and integrins (PubMed:35150743), which may contribute to tropism for olfactory epithelial cells. Related references: PMID:33082294 PMID:33082293 PMID:35150743
- After receptor binding, the S protein undergoes proteolytic cleavage by host TMPRSS2 or cathepsin L to trigger membrane fusion, acting as a class I viral fusion protein (PubMed:32142651, PubMed:32703818, PubMed:34159616, PubMed:34561887). Related references: PMID:32142651 PMID:32703818 PMID:34159616 PMID:34561887
- The spike precursor is processed into S1 and S2 subunits; the S2 subunit contains fusion peptides and heptad repeats that refold to drive viral envelope–host membrane merger.
- Subcellularly, the spike protein localizes to the virion membrane, host endoplasmic reticulum–Golgi intermediate compartment, and host cell membrane.
- The stalk domain of S contains three flexible hinges, granting the receptor-binding head considerable orientational freedom (PubMed:32817270). Related references: PMID:32817270
Experimental Guidance and Technical Tips
- The immunogen is a recombinant NSP7 protein fragment; for Western blot, consider validating detection of endogenous NSP7 in SARS-CoV-2–infected cell lysates or using recombinant NSP7 as a positive control.
- As a polyclonal antibody, lot-to-lot variation may occur; include appropriate negative controls and optimize dilution factors for each new lot.
- For ELISA, pairing with a validated NSP7 detection antibody or antigen may improve assay sensitivity; perform checkerboard titrations to determine optimal concentrations.
- Note that the predicted molecular weight of NSP7 is approximately 9.6 kDa; if unexpected bands are observed, evaluate sample preparation and antibody specificity.
