Biological Background: SARS-CoV-2 NSP3 Function and Localization
- SARS-CoV-2 NSP3 is a component of the replicase polyprotein 1ab (also known as ORF1ab polyprotein) that is cleaved by its own papain-like protease activity to release the NSP3 protein.
- NSP3 possesses papain-like protease (PL-PRO) activity responsible for cleavages at the N-terminus of the polyprotein, and collaborates with NSP4 to assemble double-membrane vesicles (DMVs) for viral replication (PubMed:35551511). Related references: PMID:35551511
- It antagonizes innate immune induction of type I interferon by blocking phosphorylation, dimerization, and nuclear translocation of host IRF3 (PubMed:32733001), and prevents NF-kappa-B signaling (By similarity). Related references: PMID:32733001
- NSP3 acts as a deubiquitinating and deISGylating enzyme, processing both Lys-48- and Lys-63-linked polyubiquitin chains, and preferentially removing ISG15 from IFIH1/MDA5 (PubMed:32726803, PubMed:33727702). Related references: PMID:32726803, PMID:33727702
- The protein exhibits ADP-ribose hydrolase activity, removing mono-ADP-ribose from host proteins, likely interfering with host PARP-mediated immune defenses (Probable) (PubMed:37242344). Related references: PMID:37242344
- Subcellular localization includes host cytoplasm, endoplasmic reticulum membrane, and double-membrane vesicles, consistent with its role in replication organelle formation.
- Keywords include viral immunoevasion, inhibition of host innate immune response, and deubiquitinase activity, highlighting its multifunctional role in host-virus interaction.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1-200 of NSP3 (YP_009725299.1), a region within the N-terminal domain; this may capture epitopes critical to its protease or ADP-ribose binding functions.
- For Western blot (WB), consider using whole-cell lysates from SARS-CoV-2 infected cells, but validate the expected band (approximately 794 kDa for the full polyprotein or cleaved NSP3 fragment) in your specific system.
- ELISA applications can be developed using this polyclonal antibody as capture or detection reagent; pairing with a monoclonal antibody may improve specificity.
- Cross-reactivity has been confirmed with SARS-CoV-2; testing with other coronaviruses is recommended if needed.
