Biological Background: NSP15 Function and Localization

• NSP15 is encoded by the rep gene of SARS-CoV-2 as part of the large ORF1ab polyprotein (also known as replicase polyprotein 1ab), which is proteolytically processed into multiple non-structural proteins (NSPs) essential for the viral life cycle.
• NSP15 functions as a uridylate-specific endoribonuclease (EndoU) that plays a pivotal role in viral transcription and replication by degrading the 5'-polyuridines generated during replication of the poly(A) region of viral genomic and subgenomic RNAs. This activity prevents the accumulation of poly(U) RNA that would otherwise hybridize with poly(A) tails and activate host dsRNA sensors, thereby avoiding innate immune detection. • The endonuclease activity of NSP15 prevents the simultaneous activation of host cell dsRNA sensors, including MDA5/IFIH1, OAS, and PKR, facilitating immune evasion.

For Western blot (WB), consider using lysates from SARS-CoV-2 infected cells and validate the band pattern with appropriate controls.
• For ELISA, immobilize recombinant NSP15 protein or the immunizing peptide, and optimize coating conditions and antibody dilutions. Since this is a polyclonal antibody, batch-to-batch consistency should be verified.
