Biological Background: Spike glycoprotein Function and Localization
- The spike glycoprotein (S) is a trimeric class I fusion protein that mediates viral entry by binding to the host receptor angiotensin-converting enzyme 2 (ACE2) (PubMed:32142651, PubMed:32155444, PubMed:33607086). Related references: PMID:32142651 PMID:32155444 PMID:33607086
- Upon receptor binding, the S protein undergoes proteolytic cleavage at the S2' site by host proteases such as TMPRSS2 or cathepsin L, triggering irreversible conformational changes that drive membrane fusion (PubMed:32142651, PubMed:32703818, PubMed:34159616, PubMed:34561887). - The spike protein is also capable of using alternative receptors for entry, including neuropilin-1 (NRP1) and neuropilin-2 (NRP2), which may contribute to infection of olfactory epithelial cells with low ACE2 expression (PubMed:33082294, PubMed:33082293). Experimental Guidance and Technical Tips
- For Western blot (WB), a band at ~34 kDa (3CLpro monomer) may be observed; consider using protease inhibitors during sample preparation to prevent degradation of the viral protease.
- In immunofluorescence (IF/ICC), ensure proper fixation and permeabilization for detection of cytoplasmic 3CLpro in SARS-CoV-2-infected cells; validate in appropriate positive-control cell lines.
- ELISA applications may require titration of the antibody and recombinant 3CLpro antigen; consider a sandwich ELISA format for high sensitivity.
- Due to sequence conservation, cross-reactivity with 3CLpro from other coronaviruses cannot be excluded; verify specificity in the relevant sample system.
