Biological Background: Sarm1 Function and Localization

  • SARM1 (Sterile alpha and TIR motif-containing protein 1), also known as NAD(+) hydrolase SARM1, is encoded by the Sarm1 gene (Kiaa0524) and undergoes alternative splicing. The protein functions primarily as an NAD(+) hydrolase, catalyzing the cleavage of NAD(+) into ADP-D-ribose (ADPR), cyclic ADPR (cADPR), and nicotinamide, thereby regulating NAD(+) metabolism and playing a pivotal role in axonal degeneration following injury.   • It acts as a negative regulator of MYD88- and TRIF-dependent toll-like receptor (TLR) signaling by promoting Wallerian degeneration—an injury-induced programmed subcellular death involving the degeneration of the axon distal to the injury site.
