- Sarcospan (SSPN), also known as K-ras oncogene-associated protein, is a transmembrane protein and a component of the dystrophin-glycoprotein complex (DGC).
- Within the DGC, sarcospan preferentially associates with the sarcoglycan subcomplex, contributing to the mechanical linkage between the F-actin cytoskeleton and the extracellular matrix in muscle cells.
- The SSPN gene undergoes alternative splicing, giving rise to multiple isoforms. Isoform 1 exhibits exclusive expression in heart and skeletal muscle, whereas isoform 2 shows broader tissue distribution including thymus, prostate, testis, ovary, small intestine, colon, and spleen.
- Subcellularly, sarcospan localizes to the cell membrane and sarcolemma, with enrichment at the postsynaptic membrane of the neuromuscular junction, highlighting its role in synaptic architecture.
