Biological Background: SAR1B Function and Localization

  • SAR1B (Secretion-associated Ras-related GTPase 1B) is a small GTPase of the Arf family that cycles between an active GTP-bound and inactive GDP-bound state.
  • Active GTP-bound SAR1B inserts into the endoplasmic reticulum (ER) membrane and recruits the COPII coat complex to mediate vesicle budding for ER-to-Golgi transport.

  • SAR1B specifically interacts with the cargo receptor SURF4 to mediate the transport of lipid-carrying lipoproteins such as APOB and APOA1 from the ER to the Golgi, thereby regulating lipid homeostasis.

  • In addition to its vesicle trafficking role, SAR1B acts as a leucine sensor: in the absence of leucine, it binds the GATOR2 complex via MIOS to inhibit TORC1 signaling, a function independent of its GTPase activity.
