Biological Background: S-Arrestin Function and Localization

S-arrestin (also known as 48 kDa protein, retinal S-antigen, rod photoreceptor arrestin) is encoded by the SAG gene in humans.

It binds to photoactivated, phosphorylated rhodopsin (RHO) and terminates RHO signaling by competing with transducin for the same binding site on RHO.

This function is critical for preventing light-dependent degeneration of retinal photoreceptor cells. Related references: PMID:9565049

S-arrestin is a member of the arrestin family, which regulates G protein-coupled receptor (GPCR) signaling.

Subcellularly, it localizes to the photoreceptor outer segment, specifically to the proximal portion of rod outer segments, and is associated with the cell membrane and cilium.

The protein is phosphorylated (phosphoprotein) and has a molecular weight of approximately 45 kDa.

Tissue expression is predominantly in the retina, especially in rod photoreceptor cells.

For Western blotting (WB), an expected band around 45 kDa should be observed; consider using retinal or photoreceptor-derived samples as positive control.

In immunohistochemistry (IHC-P), SAG expression is predominantly in the retina; validate with appropriate tissue sections.
