Biological Background: S-Arrestin Function and Localization

  • S-arrestin (also known as 48 kDa protein, retinal S-antigen, rod photoreceptor arrestin) is encoded by the SAG gene in humans.

  • It binds to photoactivated, phosphorylated rhodopsin (RHO) and terminates RHO signaling by competing with transducin for the same binding site on RHO.

  • This function is critical for preventing light-dependent degeneration of retinal photoreceptor cells. Related references: PMID:9565049

  • S-arrestin is a member of the arrestin family, which regulates G protein-coupled receptor (GPCR) signaling.

  • Subcellularly, it localizes to the photoreceptor outer segment, specifically to the proximal portion of rod outer segments, and is associated with the cell membrane and cilium.

  • The protein is phosphorylated (phosphoprotein) and has a molecular weight of approximately 45 kDa.

  • Tissue expression is predominantly in the retina, especially in rod photoreceptor cells.

  • Mutations in SAG are associated with ocular diseases such as congenital stationary night blindness and retinitis pigmentosa.

Experimental Guidance and Technical Tips

  • The immunogen used is a recombinant protein spanning amino acids 1–405 of human SAG (full-length), which may recognize the native protein across multiple applications.

  • For Western blotting (WB), an expected band around 45 kDa should be observed; consider using retinal or photoreceptor-derived samples as positive control.

  • In immunohistochemistry (IHC-P), SAG expression is predominantly in the retina; validate with appropriate tissue sections.

  • The antibody cross-reacts with mouse and rat SAG, enabling its use in rodent models; however, confirm reactivity experimentally.

  • For ELISA, the antibody may detect recombinant or native S-antigen; optimize coating and detection conditions.
