Biological Background: SAFB Function and Localization
- SAFB (Scaffold attachment factor B1), also known as HSP27 ERE-TATA-binding protein, HAP, HET, or SAFB1, is an alternative-spliced nuclear protein that functions as an estrogen receptor corepressor and DNA/RNA-binding factor.
- It binds to scaffold/matrix attachment region (S/MAR) DNA and assembles a 'transcriptosomal' complex containing SR proteins and RNA polymerase II, coupling transcription and RNA processing (PubMed:9671816). Related references: PMID:9671816
- SAFB acts as an estrogen receptor corepressor and can bind to the HSP27 promoter to decrease its transcription, thereby negatively regulating cell proliferation (PubMed:12660241). Related references: PMID:12660241
- When associated with RBMX, SAFB binds and stimulates transcription from the SREBF1 promoter (By similarity).
- Subcellular localization is nuclear.
- It is ubiquitously expressed, with high levels in the central nervous system and low levels in the liver; also expressed in many breast cancer cell lines.
- Post-translational modifications include acetylation, methylation, phosphorylation, and ubiquitin conjugation (isopeptide bond), as indicated by keywords.
Experimental Guidance and Technical Tips
- The immunogen used is a recombinant fusion protein spanning the C‑terminal region (amino acids 815‑915) of human SAFB. This region may contribute to antibody specificity; cross‑reactivity with SAFB isoforms or related proteins should be evaluated in the relevant sample system.
- For Western blot, SAFB has a predicted molecular weight of ~103 kDa; consider using a low‑percentage (e.g., 8%) SDS‑PAGE gel and appropriate transfer conditions for high‑molecular‑weight proteins.
- For immunohistochemistry (IHC‑P), optimal antigen retrieval and detection conditions may vary. Validate the antibody in known positive tissues such as human CNS sections or breast cancer cell lines.
- In ELISA applications, determine optimal coating antigen and antibody dilutions empirically.
