Biological Background: SAA1 Function and Localization
- SAA1 (Serum Amyloid A1) is a major acute phase protein whose expression is dramatically upregulated during inflammation. It is primarily synthesized by the liver and secreted into the plasma.
- The protein is produced as a preprotein with an N-terminal signal peptide, which is cleaved to yield the mature secreted form of approximately 12 kDa.
- SAA1 circulates in the blood predominantly associated with high-density lipoprotein (HDL) and can also bind heparin, influencing lipid metabolism and immune responses.
- Post-translational methylation has been identified on SAA1, which may regulate its stability or function.
- Under chronic inflammatory conditions, SAA1 can undergo proteolytic processing and misfolding, leading to the formation of amyloid fibrils and contributing to secondary (AA) amyloidosis.
- Human SAA1 is a member of the serum amyloid A family, which includes acute phase isoforms SAA1 and SAA2, sharing high sequence homology.
- Keywords from UniProt indicate involvement in amyloidosis, acute phase response, and reference proteome categories.
Experimental Guidance and Technical Tips
- Immunogen design: The antibody was generated against a synthetic peptide within the N-terminal region (amino acids 1-100) of human SAA1. This region is conserved across human, mouse, and rat, consistent with the reported cross-reactivity.
- For Western blot (WB): A band at approximately 12 kDa is expected for endogenous SAA1. Consider using positive controls such as liver lysates or plasma samples from acute phase-induced models.
- ELISA applications: The antibody can be used as a capture or detection reagent in sandwich ELISA for quantifying SAA1 in biological fluids. Pairing with another clone may be necessary; optimization is recommended.
- Due to its polyclonal nature, the antibody may detect multiple epitopes or low-level background; titration and appropriate blocking are advised.
- Although validated for human, mouse, and rat, reactivity in other species should be tested individually.
