Biological Background: S1PR3 Function and Localization

  • Sphingosine 1-phosphate receptor 3 (S1PR3), also designated as EDG3, LPB3, S1P3, or EDG-3, is a member of the G protein-coupled receptor family.
  • It serves as a high-affinity receptor for the lysosphingolipid sphingosine 1-phosphate (S1P), a bioactive lipid mediator involved in numerous cellular processes.
  • In rat hepatoma cells, S1PR3 has been shown to mediate S1P-induced cell proliferation and suppression of apoptosis, highlighting its role in cell survival signaling.
  • S1PR3 localizes primarily to the cell membrane, consistent with its function as a transmembrane receptor.
  • It is widely expressed across human tissues, with particularly high expression levels observed in the heart, placenta, kidney, and liver.
  • The receptor undergoes post-translational modifications, including glycosylation and phosphorylation, and features multiple transmembrane helices characteristic of GPCRs.

Experimental Guidance and Technical Tips

  • Because the immunogen corresponds to an N-terminal sequence, the antibody may preferentially recognize the native, unmodified form; performance on denatured samples (WB) should be verified.
  • For Western blot, the predicted molecular weight is approximately 42 kDa, but glycosylation or other modifications may cause a shift; consider this during band identification.
  • For ELISA, optimization of coating antigen and antibody concentration is recommended, especially when using recombinant protein or cell lysates.
  • Cross-reactivity with mouse and rat S1PR3 has been observed; however, sequence homology should be checked for other species if needed.
  • Always validate in the relevant sample system, as post-translational modifications or expression levels may vary across tissues and cell lines.
