Biological Background: S100B Function and Localization
- S100B (S-100 protein beta chain) is a small zinc- and calcium-binding protein highly expressed in astrocytes, where it constitutes one of the most abundant soluble brain proteins.
Related references: PMID:20950652 PMID:6487634 - The protein weakly binds calcium but tightly binds zinc, with distinct binding sites for each ion on each monomer; physiological potassium concentrations antagonize both divalent cation binding.
Related references: PMID:20950652 PMID:6487634 - S100B acts as a neurotrophic factor, promoting astrocytosis and axonal proliferation, and is also involved in sympathetic innervation of thermogenic adipose tissue as an adipocyte-derived neurotrophic factor.
- It may assist ATAD3A cytoplasmic processing, preventing aggregation and favoring mitochondrial localization.
Related references: PMID:20351179 - S100B can mediate calcium-dependent regulation by interacting with TPR-containing proteins and modulating their activity.
Related references: PMID:22399290 - Post-myocardial infarction, interaction with AGER may trigger myocyte apoptosis via ERK1/2 and p53/TP53 signaling.
- Subcellular localization includes cytoplasm, nucleus, and secretion.
- Although predominantly a brain protein, S100B is also detected in a variety of other tissues.
Experimental Guidance and Technical Tips
- The immunogen corresponds to full-length human S100B (amino acids 1-92), which is highly conserved across human, mouse, and rat, supporting the reported cross-reactivity.
- For Western blotting, use a low molecular weight marker and optimize transfer conditions given the ~11 kDa protein size.
- For immunohistochemistry (IHC-P), consider citrate-based antigen retrieval and validate in the relevant tissue type; for immunofluorescence (IF-P), include appropriate permeabilization steps.
