Biological Background: S100A8 Function and Localization
- S100A8 (Protein S100-A8), also known as calgranulin-A, MRP-8, or CFAG, is a calcium- and zinc-binding protein and a member of the S100 family of EF-hand calcium-binding proteins.
- It forms the calprotectin heterodimer (S100A8/A9), which plays a central role in regulating inflammatory processes and immune responses, inducing neutrophil chemotaxis and adhesion.
- Intracellularly, calprotectin facilitates leukocyte arachidonic acid trafficking, modulates the tubulin-dependent cytoskeleton, and activates the neutrophilic NADPH-oxidase, contributing to microbial killing.

- Extracellularly, S100A8 acts as an alarmin/DAMP, binding to TLR4 and AGER to activate MAPK and NF-kB pathways, amplifying pro-inflammatory cascades, and exerts antimicrobial activity via Zn2+ chelation.
- The protein is localized in the cytoplasm, cytoskeleton, cell membrane, and can be secreted; it is predominantly expressed in myeloid cells and strongly upregulated in inflammatory conditions and various tumors.
- S100A8 is implicated in diverse processes including oxidative stress scavenging, apoptosis, autophagy, and S-nitrosylation, as highlighted by UniProt keywords.
Experimental Guidance and Technical Tips
- The immunogen comprises amino acids 1–93 of human and mouse S100A8, covering the full-length protein; thus, the antibody is expected to recognize both species with high specificity.
- For Western blot (WB), the predicted molecular weight is approximately 10 kDa for human and 11 kDa for mouse; consider using high-percentage gels to resolve low-molecular-weight proteins.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF-P), appropriate antigen retrieval and permeabilization steps should be optimized, as S100A8 localizes to cytoplasm and can be secreted.
- For ELISA, the antibody may be used as a capture or detection reagent; validate performance in your specific assay system with appropriate controls.
