  • S100A8 is a small calcium- and zinc-binding protein (10295 Da) belonging to the S100 family, also known as calgranulin-A, MRP-8, p8, and CP-10.
  • It plays a prominent role in the regulation of inflammatory processes and innate immune responses, primarily by inducing neutrophil chemotaxis and adhesion.
  • Intracellularly, as part of the S100A8/A9 (calprotectin) complex, it facilitates leukocyte arachidonic acid trafficking, modulates tubulin-dependent cytoskeleton reorganization during phagocyte migration, and activates the NADPH-oxidase by promoting enzyme complex assembly and transferring arachidonic acid to the complex.

Experimental Guidance and Technical Tips

  • The immunogen used was a recombinant protein covering amino acids 1-89 of mouse S100A8 (NP_038678.1). Given this sequence corresponds to nearly the full-length protein (predicted molecular weight ~10 kDa), the antibody should recognize the native form. For Western blot, expect a band around 10–12 kDa; reduction and denaturation may help expose the epitope fully.
  • As a polyclonal antibody, it may have slight lot-to-lot variation; consider validating performance with a known positive control such as mouse bone marrow or spleen lysate, or cells stimulated to express S100A8 (e.g., LPS-treated macrophages).
  • For immunohistochemistry (IHC-P), antigen retrieval conditions (e.g., citrate buffer pH 6.0 heat-induced epitope retrieval) should be optimized, and endogenous peroxidase quenching may be necessary. Check cross-reactivity with rat tissues as noted.
  • In ELISA applications, coat recombinant S100A8 or use a sandwich format with a matched antibody pair; confirm specificity with relevant negative controls.
