Biological Background: S100A8 Function and Localization
- Protein S100-A8, also known as calgranulin-A or MRP8, is a small calcium- and zinc-binding protein of the S100 family.
- It forms a heterocomplex with S100A9 known as calprotectin, which exerts pleiotropic intracellular and extracellular functions in inflammation and immune response.
- Intracellularly, it facilitates arachidonic acid trafficking, modulates cytoskeleton during phagocyte migration, and activates NADPH-oxidase.
- Participates in regulatory T-cell differentiation together with CD69. Related references: PMID:26296369
- Extracellular S100A8 acts as a DAMP/alarmin, binding TLR4 and AGER to amplify pro-inflammatory cascades.
- Possesses antimicrobial activity against bacteria and fungi likely via Zn2+ chelation.
- Predominantly expressed in myeloid cells (neutrophils and monocytes), strongly upregulated in inflammatory conditions and many tumors.
- Subcellularly found in cytoplasm, cytoskeleton, cell membrane, and secreted.
