Biological Background: S100A8 Function and Localization

  • Protein S100-A8, also known as calgranulin-A or MRP8, is a small calcium- and zinc-binding protein of the S100 family.
  • It forms a heterocomplex with S100A9 known as calprotectin, which exerts pleiotropic intracellular and extracellular functions in inflammation and immune response.
  • Intracellularly, it facilitates arachidonic acid trafficking, modulates cytoskeleton during phagocyte migration, and activates NADPH-oxidase.

  • Extracellular S100A8 acts as a DAMP/alarmin, binding TLR4 and AGER to amplify pro-inflammatory cascades.
  • Possesses antimicrobial activity against bacteria and fungi likely via Zn2+ chelation.
