  • S100A8, also known as Calgranulin-A, MRP8, or CFAG, is a calcium- and zinc-binding protein of the S100 family, predominantly expressed in myeloid cells such as neutrophils and monocytes.
  • Intracellularly, as part of the calprotectin heterodimer (S100A8/A9), it facilitates arachidonic acid trafficking, modulates cytoskeleton dynamics during phagocyte migration, and activates the neutrophilic NADPH-oxidase by aiding enzyme complex assembly and transferring arachidonic acid.
  • Extracellularly, S100A8 acts as a pro-inflammatory alarmin/DAMP, recruiting leukocytes and promoting cytokine/chemokine production by binding to TLR4 and AGER, thereby amplifying inflammatory cascades via MAPK and NF-kB pathways.
  • It exhibits antimicrobial activity against bacteria and fungi likely through Zn²⁺ chelation, and can induce autophagy and apoptosis via ROS and BNIP3.
