Biological Background: S100A8 Function and Localization

  • Protein S100-A8, also known as calgranulin-A, MRP-8, or S100 calcium-binding protein A8, is a member of the S100 family of calcium-binding proteins encoded by the S100A8 gene in humans.
  • S100A8 forms heterodimers with S100A9 to constitute calprotectin, which plays a key role in regulating inflammatory processes and immune responses, including neutrophil chemotaxis and adhesion.
  • Intracellularly, S100A8 facilitates leukocyte arachidonic acid trafficking, modulates the cytoskeleton during phagocyte migration, and activates the NADPH-oxidase by aiding assembly of the enzyme complex at the cell membrane.
  • Extracellularly, S100A8 acts as an alarmin or danger-associated molecular pattern (DAMP), stimulating innate immune cells via pattern recognition receptors such as TLR4 and AGER, and activating MAP-kinase and NF-kappa-B pathways to amplify pro-inflammatory cascades.
  • S100A8 exhibits antimicrobial activity toward bacteria and fungi, likely through Zn2+ chelation, and can induce cell death via autophagy and apoptosis involving BNIP3 and reactive oxygen species.   • Subcellular localization includes secretion, cytoplasm, cytoskeleton, and cell membrane. Tissue expression is predominantly in myeloid cells, specifically circulating neutrophils and monocytes, and is strongly upregulated under chronic inflammatory conditions and in many tumors.
  • Post-translational modifications, such as S-nitrosylation at cysteine residues, modulate its functions, including selective S-nitrosylation of target proteins via the iNOS-S100A8/A9 transnitrosylase complex.

Experimental Guidance and Technical Tips

  • The immunogen covers nearly full-length human S100A8 (amino acids 1–93); therefore, the antibody may recognize both monomeric and calprotectin-complexed forms.
  • For Western blot (WB), expect a band at approximately 11 kDa; validate with appropriate positive controls such as neutrophil or monocyte lysates.
  • In immunohistochemistry (IHC-P), consider antigen retrieval methods and include myeloid-rich tissues (e.g., inflamed or tumor samples) as positive controls.
  • When using immunofluorescence (IF/ICC), S100A8 may localize to the cytoplasm, cell membrane, or be secreted, so fixation and permeabilization conditions should be optimized.
  • This antibody is reported to cross-react with mouse and rat; however, independent validation in your specific experimental system is recommended.
