Biological Background: S100A16 Function and Localization
- S100A16 (Protein S100-A16) is a member of the S100 calcium-binding protein family, encoded by the S100A16 gene (also known as S100F).
- S100A16 binds one calcium ion per monomer with high affinity, functioning as a calcium sensor. Related references: PMID:17030513
- It is involved in adipocyte biology: it can promote adipocyte differentiation in vitro, and its overexpression in preadipocytes enhances proliferation and adipogenesis while reducing insulin-stimulated glucose uptake.
- The protein localizes to the nucleus, nucleolus, and cytoplasm, suggesting roles in both nuclear and cytoplasmic processes.
- S100A16 exhibits ubiquitous tissue expression, with highest levels in esophagus, adipose tissue, and colon; lower expression in lung, brain, pancreas, and skeletal muscle. Expression is upregulated in several tumor types (bladder, lung, thyroid, pancreas, ovary) and is found in astrocytes. Related references: PMID:14684152 PMID:17030513
- The protein contains calcium-binding EF-hand repeats and also binds zinc, as indicated by proteomics and structural studies.
Experimental Guidance and Technical Tips
- For Western blotting, the predicted molecular weight of S100A16 is approximately 12 kDa; use a high-percentage SDS-PAGE gel (e.g., 15%) and low-molecular-weight markers for optimal resolution.
- Given the nuclear and cytoplasmic localization of S100A16, consider subcellular fractionation to enrich target protein in complex samples.
- Test the antibody in relevant human, mouse, or rat cell lines or tissues known to express S100A16, such as adipocytes, esophageal epithelium, or tumor lines.
- For ELISA, titrate the antibody as a capture or detection reagent and validate with recombinant S100A16 or native protein in lysates; pair with a matched detection antibody for sandwich ELISA if needed.
- Since S100A16 is a calcium-binding protein, include calcium-chelating agents (e.g., EDTA) in lysis buffers with caution, as they may affect protein conformation.
