• Protein S100-A12, also known as calgranulin-C, EN-RAGE, and MRP-6, is a member of the S100 family of calcium-binding proteins.
• It functions as a pro-inflammatory alarmin or DAMP, activating innate immune responses via the receptor for advanced glycation endproducts (RAGE/AGER).
• Upon AGER binding, it triggers MAP-kinase and NF-κB signaling, leading to cytokine production and upregulation of adhesion molecules such as ICAM1 and VCAM1.
• It acts as a chemoattractant for monocytes and mast cells, and can stimulate mast cell degranulation.
