- Protein S100-A12, also known as calgranulin-C, EN-RAGE, and MRP-6, is a member of the S100 family of calcium-binding proteins.
- It functions as a pro-inflammatory alarmin or DAMP, activating innate immune responses via the receptor for advanced glycation endproducts (RAGE/AGER).
- Upon AGER binding, it triggers MAP-kinase and NF-κB signaling, leading to cytokine production and upregulation of adhesion molecules such as ICAM1 and VCAM1.
- It acts as a chemoattractant for monocytes and mast cells, and can stimulate mast cell degranulation.
- S100A12 exhibits antimicrobial activity; the calcitermin fragment has antifungal (C. albicans) and antibacterial (E. coli, P. aeruginosa) properties.
- It is predominantly expressed in neutrophils, monocytes, and activated macrophages, and is secreted into extracellular spaces.
- Subcellularly, it is found in the cytoplasm, associated with the cytoskeleton, and at the cell membrane.
- The protein binds calcium, zinc, and copper; it can inhibit matrix metalloproteinases (MMP2, MMP3, MMP9) by chelating zinc.
