Biological Background: S100A12 Function and Localization
- S100A12 is a calcium-, zinc- and copper-binding protein of the S100 family, also known as EN-RAGE, calgranulin-C (CAGC), MRP-6, and p6.
- It plays a prominent role in regulating inflammatory processes and immune response by acting as a DAMP/alarmin molecule.
- Upon binding to RAGE (AGER), S100A12 activates MAP-kinase and NF-kappa-B pathways, inducing pro-inflammatory cytokines and upregulating adhesion molecules ICAM1 and VCAM1.
- It chemoattracts monocytes and mast cells, stimulates mast cell degranulation, and promotes leukocyte recruitment to sites of inflammation.
- S100A12 inhibits matrix metalloproteinases (MMP2, MMP3, MMP9) by chelating zinc from their active sites.
- It has antimicrobial properties, including antifungal activity against C. albicans and activity against E. coli and P. aeruginosa.
- Predominantly expressed in neutrophils, monocytes, activated macrophages, and eosinophils in asthmatic airways; elevated serum levels are associated with inflammatory disorders.
- Subcellular localization includes secreted, cytoplasmic, cytoskeletal, and cell membrane compartments.
