Biological Background: S100A12 Function and Localization
- Protein S100-A12, also known as calgranulin-C, EN-RAGE (Extracellular Newly Identified RAGE-binding Protein), CAAF1, MRP-6, and p6, is a member of the S100 calcium-binding protein family.
- It plays a central role in inflammatory and immune responses by functioning as a danger-associated molecular pattern (DAMP) molecule, binding to the receptor for advanced glycation endproducts (AGER/RAGE) and activating MAP-kinase and NF-kappa-B pathways to promote cytokine production and up-regulate adhesion molecules like ICAM1 and VCAM1.
- Acts as a chemoattractant for monocytes and mast cells, and can stimulate mast cell degranulation, amplifying leukocyte recruitment to inflammation sites.
- Exhibits calcium-, zinc-, and copper-binding properties; it inhibits matrix metalloproteinases (MMP2, MMP3, MMP9) by chelating zinc from their active sites.
- Possesses antimicrobial activities: displays filariacidal and filariastatic effects, and its derivative calcitermin has antifungal activity against Candida albicans and antibacterial activity against E. coli and P. aeruginosa.
- Predominantly expressed in neutrophils, monocytes, and activated macrophages; elevated serum levels are observed in inflammatory disorders such as rheumatoid arthritis, Crohn's disease, and Kawasaki disease.
