Biological Background: Retinoic Acid Receptor RXR-beta Function and Localization

  • Retinoic acid receptor RXR-beta, encoded by RXRB, is also known as nuclear receptor subfamily 2 group B member 2, NR2B2, RXRbeta, and retinoid X receptor beta. It is a member of the nuclear receptor family involved in ligand-dependent transcriptional regulation.
  • RXR-beta functions as a receptor for retinoic acid, forming heterodimers with retinoic acid receptors (RARs) that bind to retinoic acid response elements (RARE) to modulate gene expression in various biological processes.
  • The RAR/RXR heterodimers are key transcriptional regulators that respond to all-trans or 9-cis retinoic acid, playing roles in development, differentiation, and homeostasis.
  • The protein localizes to both the nucleus and cytoplasm, consistent with its dynamic role in signal transduction and gene regulation.
  • RXR-beta is expressed in aortic endothelial cells (at protein level) and in monocytes, as well as in a variety of tumor cell lines. Related references: PMID:28167758 PMID:26463675
  • Structurally, it contains zinc-finger DNA-binding motifs and is subject to alternative splicing and methylation, which may affect its function and stability.
  • The calculated molecular weight is approximately 57 kDa (UniProt: 56922 Da), consistent with the reported protein weight.

Experimental Guidance and Technical Tips

  • The immunogen used is a synthetic peptide corresponding to amino acids 200-300 of human RXRB (NP_001257330.1). This region may provide specificity for RXRB; however, due to the polyclonal nature, cross-reaction with related isoforms or family members should be considered.
  • For Western blotting, the predicted protein weight is 57 kDa. Validate the antibody in human, mouse, and rat samples as cross-reactivity has been indicated, but expression levels may vary across tissues.
  • In ELISA applications, optimize coating and detection conditions, as no specific titer or working dilution is supplied. Pair with appropriate secondary antibodies for detection.
  • Consider that the protein can localize to both the nucleus and cytoplasm; thus, sample preparation for whole-cell lysates is recommended for comprehensive detection.
