Biological Background: RUNX1T1 Function and Localization
- RUNX1T1, also known as ETO/MTG8/CBFA2T1, is a transcriptional corepressor that facilitates gene silencing via DNA-binding transcription factors and recruitment of histone-modifying enzymes. Related references: PMID:10688654, PMID:12559562, PMID:15203199
- It can repress the expression of MMP7 in a ZBTB33-dependent manner. Related references: PMID:23251453
- RUNX1T1 inhibits transactivation mediated by TCF12. Related references: PMID:16803958
- It acts as a negative regulator of adipogenesis.
- The AML1-MTG8/ETO fusion protein, resulting from the t(8;21) chromosomal rearrangement, is frequently found in acute myeloid leukemia and contributes to leukemogenesis by enhancing hematopoietic stem/progenitor cell self-renewal. Related references: PMID:23812588
- RUNX1T1 is primarily localized to the nucleus.
- It is most abundantly expressed in brain, with lower levels in lung, heart, testis, and ovary.
- The protein contains zinc-finger MYND-type domains and is subject to phosphorylation; it is classified as a DNA-binding proto-oncogene and transcriptional repressor.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fragment covering amino acids 250–403 of human RUNX1T1 (NP_783552.1), which may recognize both wild-type and certain fusion proteins; validate in the relevant sample system.
- For Western blotting, the predicted molecular weight is approximately 68 kDa, but post-translational modifications may cause band shifts; consider using appropriate controls.
- The reported cross-reactivity with rat suggests potential utility in rodent models, but experimental verification is recommended for each new application.
- ELISA conditions (coating concentration, detection antibody dilution) should be optimized empirically; the antibody may be suitable as a capture or detection reagent depending on the pair.
