Biological Background: RUFY3 Function and Localization

  • RUFY3 (also known as RIPX, SINGAR1, ZFYVE30) is a RUN and FYVE domain-containing protein that functions as an effector of the small GTPase ARL8, playing a critical role in linking endolysosomes to the dynein-dynactin motor complex for retrograde transport along microtubules. In nonneuronal cells, this activity concentrates endolysosomes in the juxtanuclear area, while in hippocampal neurons it drives retrograde transport of endolysosomes from axons to the soma. Related references: PMID:35314674
  • The protein is implicated in neuronal polarity formation and axon growth; it may inhibit the formation of additional axons by suppressing PI3K activity in minor neuronal processes (By similarity).
  • RUFY3 participates in cytoskeletal organization by modulating the subcellular localization of FSCN1 and DBN1 at axonal growth cones and promotes the formation of F-actin-enriched protrusive structures at the cell periphery, contributing to cell migration and invasion. Related references: PMID:25766321
  • Subcellularly, RUFY3 localizes to the cytoplasm, endomembrane system, and various cell projections including invadopodia, growth cones, filopodia, and lamellipodia; it is also found in the perikaryon and lysosomes.
  • Tissue expression analysis reveals overexpression in gastric cancer cells and tissues at the protein level, suggesting a potential oncogenic role.
  • The protein undergoes alternative splicing and is regulated by phosphorylation; it is classified under keywords including cell junction, developmental protein, neurogenesis, and oncogene.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (amino acids 1-65) of human RUFY3; this region may be conserved across species, supporting observed cross-reactivity with mouse and rat, but experimental validation in the target species is advised.
  • For Western blot, a band around 53 kDa is expected based on the calculated molecular weight; consider using appropriate positive controls (e.g., human neuronal or cancer cell lines) and verify specificity, especially given potential isoforms from alternative splicing.
  • In ELISA applications, optimize coating antigen and detection conditions to ensure linearity and sensitivity; the recombinant immunogen can serve as a standard for assay calibration.
  • When studying RUFY3 in neuronal systems, consider that its activity is coupled to GTP-bound ARL8; functional assays may require co-expression or stimulation of endolysosomal transport pathways.
