Biological Background: RTN4RL1 Function and Localization

  • RTN4RL1 (Reticulon-4 receptor-like 1), also known as Nogo receptor-like 2 (NGRH2) or NgR3, is a cell surface receptor of the Nogo receptor family.
  • Plays a functionally redundant role in postnatal brain development and in regulating axon regeneration in the adult central nervous system; contributes to normal axon migration across the brain midline and formation of the corpus callosum.
  • Inhibits neurite outgrowth and axon regeneration by binding neuronal chondroitin sulfate proteoglycans; binds heparin (By similarity).
  • Restricts the number of dendritic spines and synapses during brain development; signaling mediates activation of Rho and downstream actin cytoskeleton reorganization (PubMed:22325200).
    Related references: PMID:22325200
  • Protects motoneurons against apoptosis, likely mediated by MAG (By similarity).
  • Localizes to the cell membrane, membrane rafts, perikaryon, and cell projections; anchored via a GPI anchor.
  • Predominantly expressed in brain, with lower levels in kidney, lung, mammary gland, placenta, salivary gland, skeletal muscle, and spleen.
  • Contains leucine-rich repeat domains and is a glycoprotein; member of the Nogo receptor family.

Experimental Guidance and Technical Tips

  • For Western blotting, use standard HRP-conjugated secondary antibodies and chemiluminescent detection. The predicted molecular weight is ~49 kDa, but glycosylation may cause an upward shift; validate with brain tissue or RTN4RL1-expressing cell lysates.
  • In ELISA applications, consider using the recombinant immunogen (aa 220-420) or full-length protein as the coating antigen to optimize binding kinetics.
  • Cross-reactivity with mouse and rat is indicated, making the antibody suitable for rodent model studies; however, confirm epitope conservation and specificity in the chosen species.
  • Due to sequence homology among Nogo receptor family members (e.g., RTN4R, RTN4RL2), verify specificity in your experimental system using appropriate controls.
