- RTN3, also known as HAP, ASYIP, NSPL2, NSPLII, or RTN3-A1, is a member of the reticulon family with a predicted molecular weight of approximately 113 kDa (112611 Da).
- It primarily localizes to the endoplasmic reticulum and Golgi apparatus membranes, where it regulates membrane trafficking in the early secretory pathway.
- RTN3 inhibits BACE1 activity and amyloid precursor protein (APP) processing, a pathway implicated in Alzheimer's disease.
- RTN3 promotes the formation of endoplasmic reticulum tubules. Related references: PMID:25612671
- It plays a role in apoptosis by inducing the caspase-8 cascade and favoring BCL2 translocation to mitochondria under ER stress.
- As an inflammation-resolving regulator, RTN3 interacts with TRIM25 and RIGI, impairing RIGI ubiquitination and inhibiting IRF3 and NF-kappa-B signaling.
- Isoform 1 is highly expressed in brain and low in skeletal muscle; isoform 2 is restricted to melanoma; isoform 3 is widely expressed, with highest levels in brain neuronal cell bodies.
