Biological Background: Rhotekin Function and Localization
- Rhotekin (RTKN) is a protein encoded by the RTKN gene in humans, also referred to as RTKN1. Alternative splicing generates multiple transcript variants.
- It acts as a downstream mediator of Rho GTPase signaling, activating the NF-kappa-B transcription factor, which may promote apoptosis resistance, particularly in gastric tumorigenesis.
- RTKN may also play a role in organizing septin cytoskeletal structures.
- The protein is highly expressed in prostate tissue, with moderate levels in kidney, heart, brain, spleen, testis, placenta, small intestine, pancreas, skeletal muscle, and peripheral blood leukocytes; low expression is observed in ovary, colon, and thymus.
- RTKN is weakly expressed in normal cell lines but overexpressed in various cancer cell lines, suggesting a role in oncogenesis.
- Post-translational modifications include phosphorylation and methylation, and the protein features coiled-coil and GTP-binding domains.
Experimental Guidance and Technical Tips
- For Western blotting, detect a band at approximately 63 kDa (predicted molecular weight 62.7 kDa) in relevant tissue or cell lysates; use a reduced loading buffer and appropriate controls.
- In immunofluorescence/immunocytochemistry, optimize fixation and permeabilization conditions; RTKN may localize to the cytoplasm or associate with septin filaments.
- The antibody cross-reacts with mouse and rat, so validate in the desired model system; sequence homology in the immunogen region supports this cross-species recognition.
- For ELISA, consider sandwich or indirect formats using the recombinant immunogen as a positive reference standard.
