Biological Background: RSK2 Function and Localization
- RSK2 (Ribosomal protein S6 kinase alpha-3), also known as RPS6KA3, p90RSK3, MAPKAPK1B, and ISPK-1, is a serine/threonine-protein kinase encoded by the RPS6KA3 gene on human chromosome X. It belongs to the AGC kinase family and contains two kinase domains connected by a linker region.
- Acting downstream of the ERK/MAPK signaling cascade, RSK2 directly phosphorylates transcription factor CREB1 and histone H3 at Ser-10 upon EGF stimulation, triggering immediate-early gene expression (PubMed:10436156, 9770464).

- The immunogen is a synthetic peptide corresponding to amino acids 600-700 of human RSK2, a region conserved across human, mouse, and rat, which supports the observed cross-reactivity. For Western blot, a band at approximately 84 kDa is expected; consider using reduced samples and loading 20-30 µg of cell lysate. Validate band specificity using appropriate positive controls (e.g., HeLa, A431, or NIH/3T3 cells) and negative controls (e.g., RSK2 knockout lysates or siRNA-treated samples).
- For immunohistochemistry (IHC-P), heat-mediated antigen retrieval with citrate buffer (pH 6.0) is recommended. Titrate the primary antibody starting at 1:50-1:200 dilution, and optimize detection with a polymer-based HRP system. Always include isotype controls and tissue sections known to express RSK2.
- In ELISA applications, consider coating with recombinant RSK2 protein or peptide and use a standard indirect ELISA protocol. The optimal antibody working concentration should be determined empirically by checkerboard titration.
