Biological Background: RSAD2 Function and Localization
- RSAD2 (viperin, cig5, vig1) is an interferon-inducible antiviral protein that establishes a broad cellular antiviral state in response to type I and type II interferons. Related references: PMID:31812350
- It functions as an S-adenosylmethionine-dependent nucleotide dehydratase, converting CTP to ddhCTP. This metabolite acts as a chain terminator for viral RNA-dependent RNA polymerases, directly inhibiting replication of numerous viruses including HCMV, HCV, WNV, dengue virus, influenza A, and HIV-1. Related references: PMID:29925952 PMID:30872404 PMID:30587778 PMID:30934824
- RSAD2 potentiates innate immune signaling by facilitating Lys-63-linked ubiquitination of IRAK1 via TRAF6, thereby enhancing TLR7/9-dependent IFN-beta production in plasmacytoid dendritic cells. Related references: PMID:30872404
- In adaptive immunity, RSAD2 modulates CD4+ T-cell activation and Th2 differentiation by regulating NFKB1 and JUNB activities, thereby promoting GATA3 activation and Th2 cytokine production.
- The protein exhibits a multifaceted subcellular distribution, localizing to the endoplasmic reticulum membrane, Golgi apparatus, lipid droplets, and mitochondria (both inner and outer membranes), allowing it to interface with diverse cellular pathways.
- Structural hallmarks include a radical SAM domain with a 4Fe-4S cluster, and the protein undergoes post-translational modifications such as acetylation and ubiquitination, consistent with its roles in antiviral defense, innate immunity, and lipid metabolism.
Experimental Guidance and Technical Tips
- The immunogen spans residues 118–361, covering the core radical SAM domain, which is conserved across human, mouse, and rat, supporting the observed cross-reactivity.
- For Western blot, endogenous RSAD2 runs at ~42 kDa; however, its expression is low under basal conditions. Induction with type I interferon (e.g., IFN-α) or viral stimuli is recommended to robustly detect the protein.
- In immunofluorescence, RSAD2 can localize to the ER, Golgi, and lipid droplets; co-staining with organelle-specific markers (e.g., calnexin, GM130, BODIPY) aids in accurate interpretation.
- For ELISA, use recombinant RSAD2 protein or lysates from interferon-treated cells as a positive control. Dilutions and coating conditions must be optimized empirically.
